dc.description.abstract Sugar cane (Saccharum Officinarum) is the giant member of tropical grass family and it contains many constituents such as sucrose, fibers, water, organic acids, non organic acids,ect. Alpha hydroxy acids (AHAs) also consist in sugar cane plant and it plays remarkable roll in cosmetics industry. The Glycolic Acid is the smallest AHA, which is highly soluble in water, hydroscopic white crystal. This distinctive compound has been extensively used in formulation of personal care producís as an active ingredient and it shows number of benefits such as exfoliation of dead skin cells, improve skin texture, appearance, reduce age spots,wrinkles and fine lines, acne, slow down the aging process and ultimately smoother, fresher appearance on the skin. Project work is focused to developed method for the fractionation or isolation of Glycolic Acid from sugar cane. Major steps of methodology were sample preparation, basification, extraction, acidification, extraction, fractionation, evaporation and analysis of final extract. Sugar cane juice sample was prepared and acidified using Hydrochloric Acid. It was extracted in to organic phase that is mainly Ethyl Acetate and evaporated using rotary evaporator until dryness. TLC and HPLC method were used for the analysis of Glycolic Acid. Therefore Qualitative analysis was done using TLC using 20% MethanoI/Chloroform solvent System and Bromocresol green was used as visualizing agent. HPLC analysis was carried out in reverse phase HPLC in order to quantify Glycolic Acid under the condition such as 70% Water/Methanol as mobile phase, at a flow rate 0.5 ml/min, at a temperature 100 °C, pressure 500kgf/cm2 and UV detector at 224 nm wave length. Finally sucrose analysis was done for the extract residue. 906 mg and 1056.5 mg of Glycolic Acid were extracted from 1 liteir of sugar cane juice using Method 1 and Method 2 respectively. Thus 0.41% glycolic acid was contained in sugar ca ne juice solid. In addition to 5.89% and 4.30% sucrose amount were consisted in Method 1 and Method 2 extract respectively. en_US
